By increasing the chain length (> C6:0) of our photo-active amphiphiles, the binding of DNA was improved, though this stable binding at C14:0 hindered UV-activated DNA release. Additionally, mixing lipids of various chain lengths improved binding and release kinetics. The C6:0 lipids did not facilitate cellular uptake and transfection, while C14:0 and its mixtures enhanced transfection outcomes.
