The 2-benzazepin-3-one structure as a p-turn mimic:synthesis and conformational analysis

Turn mimetics have attracted considerable attention since beta-and y-turn conformations are frequently found in bioactive peptides.Freidinger developed gamma-,delta-and epsilon-lactams to induce turn structures in peptides.A stronger bias for P-turn formation has been obtained by using spiro-lactams.The 4-amino-l,2,4,5-tetrahydro-2-benzazepin-3-one(Aba)scaffold 1 has been used in a number of bioactive peptides.We now report the conformational analysis of Aba analogues 2-3 and the synthesis of the corresponding spiro-lactam analogue 4.
