Efficient synthesis of difficult sequence-containing peptides through O-acyl isopeptides:Application to the synthesis of Alzheimer's disease-related peptide,A beta_(1-42)

Since amyloid beta peptide(A beta)_(1-42)plays a critical role in the pathogenesis of Alzheimer's disease(AD),many studies using synthetic A beta_(1-42)have been carried out to clarify the involvement of A beta_(1-42)in AD.However,A beta_(1-42)is a highly hydrophobic peptide and forms aggregates in various media resulting in the synthetic difficulty of this peptide,so-called "difficult sequence"-containing peptide.Furthermore,the biological experiments using A beta_(1-42)are problematic because of the large extent of aggregation in a standard storage solution such as dimethylsulfoxide(DMSO).
