Phage-Library Displaying Toll-Like Receptor as Leucine-Rich Repeat (LRR) Protein Scaffold

We constructed leucine-rich repeat (LRR) protein library based on toll-like receptors (TLRs) with antigen-recognition feature of variable lymphocyte receptor (VLRs). LRR gene was composed of N-terminal capping repeat, LRR modules and C-terminal capping repeat. To confer antigen-binding specificity, LRR module was synthesized in order to randomize 5 amino acid positions and integrated into pCANTAB5E at Nhel and Avrll sites. We here examined whether this LRR gene could be expressed in Escheria coli and display on phage surface.
