In this study the fluorescence activity of a mutant form of Green Fluorescent Protein (GFP), GFPuv, was explored in ionic liquid solvents. Purified GFPuv was obtained and experiments probing the fluorescence spectral properties of GFPuv in a series of solvents were undertaken. Data show that GFPuv is fluorescent in several ionic liquid solvents when sufficient water is also present, and the amount of water (in the solvent system) is critical in determining the fluorescent activity. Results are interpreted with regards to previous Hofmeister series correlations in attempt to rationalize solvating and stabilizing properties of the solvent systems investigated.
