Comparison of Two Sample Preparation Methods for Analyzing Pain Management Drugs in Urine

1) Protein removal and depletion method is much faster and simpler than Sagacity HL SPE method. 2) Sagacity HL SPE method gave excellent recovery for all testing drugs; While Protein removal and depletion method gave relative good recovery for most analytes, but low recoveries for some very hydrophobic compounds. 3) We also evaluated both types of extraction method by DART-MS. In protein depletion and removal platefiltered samples, large quantities of other biological material were present, like creatine, which significantly interfered with the analysis and lowered the sensitivity of the method. Sagacity HL SPE method was designed to remove those very hydrophilic compounds, therefore there was very little interference from biological metabolites in the extracted samples.
