Highly Sensitive Determination of A Phosphopep tide Therapeutic in Human Plasma by LC-MS/MS

摘要

1. Plasma sample treatment with 6M guanidine (1:1) was able to significantly improve analyte stability. 2. The use of basic pH wash solution was an effective way to reduce carryover of the phosphopeptide. 3. The use of high concentration of formic acid in the mobile phases could improve chromatographic separation and peak shape. 4. The method was successfully validated over concentration range of 105000 pg/mL. Accuracy and precision have been demonstrated for this method.
