Based on the results of our investigation, polypropylene (PP) tubes were implemented for the extraction portion of the method. The assay validation with LLOQ of 0.1 ng/mL was completed successfully meeting current industry standards. Precautions have always been taken for high sensitivity assays with low LLOQ by utilizing LC/MS grade solvents, ensuring cleanliness of glassware and instrumentation, etc... Additional controls and evaluations have also been implemented in this lab to ensure the ruggedness of assays when lowering the limit of quantitation from established methods.
