This work allowed us to set up a protocol for highly purified mitochondria preparation. More than 1000 proteins and several hundred exact N-terminal sequences of mitochondrial proteins were identified. Cross-validating these identification lists with available information on known mitochondrial proteins and known and expected transit peptides cleavage sites allows interesting new observations to be made on the human mitochondrial proteome and its N-terminal processing. The data are being integrated into SwissProt and neXtProt databases what will bring all this structured knowledge on human mitochondrial proteins at the disposal of the biologists' community at large. A better characterization of the human mitochondrial proteome and its specific processing will contribute to the shift from predicted to curated protein databases, a prerequisite for the shift from high-throughput to fine characterization proteomics.
