LC/MS/MS Quantification of Kynurenine Metabolites Implicated in Neurological Disorders

摘要

1. LC/MS/MS method is capable of monitoring KYN metabolism changes in plasma, enzyme reaction buffers and microdialysis studies. 2. Preliminary results suggest the method has more then sufficient sensitivity for cell culture and brain tissue homogenate studies. 3. Use of internal standards demonstrated the presence of matrix effects in microdialysis samples which are directly injected. Implementation of a sample cleanup procedure will be tested to see if the matrix effects can be alleviated. 4. Matrix effects in plasma and brain tissue homogenates remains to be tested following sample preparation described. A similar LC/MS/MS method reported by W. Zhu et. al. indicated little sample matrix effects when cleanup of samples was performed on serum, urine and cell culture samples.
