1) The siRNA technology targeting BAX/BAK/LDH can significantly facilitate CHO cell growth by regulating target pathways. 2) In the early growth phase, day 3, significant changes in metabolic pathways caused by silencing these proteins have been found, even though the effects on the cells were not yet significantly observed. This suggests that global proteomic profiling is highly sensitive in monitoring cell culture engineering-induced changes. 3) Anti-apoptotic effect was not globally observed, as expected, likely due to the fact that (1) apoptosis had not significantly occurred as cell culture was still in the early growth phase and (2) the LDH silencing effect predominated. 4) To elucidate the effect of silencing BAX/BAK, the temporal analysis of BAX/BAK siRNA treated CHO cell samples is in progress.
