摘要

Two-stage methanol-assisted solubilization/digestion coupled with shotgun LC-MS analysis of tissue proteins in H-Ras v12 induced cardiomyopathy produced efficient phenotypic molecular profiling of a pathological process in vivo. In addition to Ras identification in the hypertrophic mouse myocardium, key proteins involved in the pathologic process under study were unambiguously identified. Subtractive set-based proteomic analysis coupled with spectral count analysis allowed discrimination of the hypertrophic myocardial phenotype from wild type as corroborated by cross-validation of selected proteins by Western blot analysis.
