We used stable isotope labeling and mass spectrometry in combination with a collection of yeast strains containing mutations in the core histones at known sites of post-translational modification, to quantitate the effect of mutation on the level of all other histone modifications. We have performed analyses on selected histone mutants and determined the existence of cross-talk between H4-K5,12 and H3-K79.
