摘要

Epitope mapping is critical in therapeutic antibody research. However, allosteric effects may occur in conventional studies which obscure the real epitopes. Time-resolved hydrogen deuterium exchange mass spectrometry (TRHDX-MS) enables the detection of millisecond timescale conformational changes in antibody-antigen (Ab-Ag) interactions. Kinetic study of an Ab-Ag complex through TRHDX-MS effectively reduces allosteric effects to give a more distinct epitope mapping.
