Imaging Mass spectrometry (IMS) is an emerging tool in proteomics, lipidomics and metabolomics. Recent developments in MALDI Imaging mass spectrometry allow today the direct detection of biomolecules such as proteins, lipids and drugs on tissue. After the demonstration of the proof of concept and publications in biomarker discovery, developments target the increase of sensitivity, image quality, and spatial resolution. It is now possible to study and image FFPE and frozen tissues, with different surface treatments. However, MS/MS identification in situ and concise proteome detection are under-developed. We used and optimized a MALDI Ion mobility Q-ToF imaging experiment on digested tissue to add an additional gas-phase-separation step in imaging mass spectrometry. This resulting data is combined with other imaging MS approaches.
