Changes in protein folding can be rapidly, efficiently and conveniently studied using Membrane ES probe pH gradients avoiding the need to run multiple sample solutions. It is well known that many proteins can undergo conformational/structural changes in solution with changes in pH and/or organic solvent composition. In the current study, a Membrane Electrospray probe was used to add charged species from a second solution flow into sample solutions through a semipermeable membrane without the direct addition of acid or organic species to the sample solution. PH scans were conducted by running acid concentration gradients in the second solution flow during ES operation. Shifts in the multiply charged ion peak profiles were observed during pH scans using the Membrane ES probe.
