Insulin secretion can be calculated by deconvolution from the measurement of C-peptide, a hormone secreted with insulin in equimolar amounts by the pancreatic beta cells. From the relationship between insulin secretion and glucose concentration a dose-response characteristic quantifying beta-cell function can be determined. This work presents a model-based method for determining simultaneously insulin secretion and the dose-response function i na subject. The method is applied to a group of 20 obese subjects who were studied during a 24 hr standardized life period. New information on the role of free fatty acids on beta-cell function is obtained.
