Conclusions: Our results demonstrated that macroporous μRB-based hydrogels are a promising novel scaffold to support osteogenesis of human MSCs. The macroporosity within μRB-based hydrogels led to accelerated new ECM deposition in a more interconnected manner with enhanced mechanical properties. The incorporation of HA into the μRB-based hydrogels allowed for significant mineralization throughout the scaffold. Our results suggest that HAnp incorporation would be a preferred method for incorporating HA into μRB-based hydrogels, and HA-coated μRB-based hydrogels can serve as a promising injectable biomaterial to enhance stem cell-based therapy for repairing large bone defects.
