Aged Decellularized Muscle Matrix Negatively Impacts Myogenic Differentiation and Fusion

Due to an increase in mortality rate with age, tissue banks have a larger supply of aged skeletal muscle tissue for decellularization. We observed an age-related loss of differentiation and myotube formation due to DMM, implying that aged DMM has a reduced myogenic potential. Our results were confirmed using protein markers for differentiation and fusion, as well as morphometric analysis. The observed increase in collagen and decrease in laminin could indicate a role for these proteins in adult myogenesis. It is possible that altering the levels of these ECM proteins could compensate the loss of myogenic potential in aged DMM. However future studies are needed to elucidate mechanisms that regulate the observed age-dependent loss of myogenic potential in DMM.
