INSOLUBILIZED DEOXYRIBONUCLEIC ACID (DNA)ABSTRACT OF THE DISCLOSUREA method of detecting antibodies to native deoxyribonucleic acid(n-DNA), anti-nuclear antibodies, antibodies to non-histone containingprotein, rheumatoid factors, antibodies to thyroglobulin, and an immuno-fluorescent assay for such detection is described. The method comprisesincubating an insoluble conjugate of methylated bovine serum albumin(mBSA)-native deoxyribonucleic acid (n-DNA) or other substrates withserum from patients with systemic lupus erythematosus (SLE) or otherautoimmune diseases for a sufficient length of time, washing the pre-cipitate or flocculate, separating and discarding therefrom the super-natant fluid, adding a fluorescein labeled anti-immunoglobulin antibodyto the washed flocculate and incubating the labeled mixture for a suf-ficient length of time, washing the incubated flocculate and separatingtherefrom the supernatant fluid, suspending the pellet in a liquid mediumto determine the fluorescence of the flocculate, which is proportionalto the concentration of antibodies to deoxyribonucleic acid and othersin serum specimen. The insoluble conjugate of methylated bovine serumalbumin (mBSA)-native deoxyribonucleic acid (n-DNA) retains the double-stranded form of native deoxyribonucleic acid (n-DNA).
