Hybrid plasmids capable of reproduction in bacillus subtilis and streptococcus pneumoniae are made by (1) characterisation of the pLS1 plasmid of S. pneumoniae and the pBD6 plasmid of b.substilis, (2) isolation and purification of both plasmids by extraction with lysozyme for b.substilis and sodium deoxicalate for s.pneumoniae, (3) digestion of both plasmids with a restriction enzyme Eco R1, (4) pptn. of the ADN in the digestion mixt. by addn. of 1 micro.l of t-RNA, 12 micro.l 3 M sodium acetate and 300 micro.l ethanol, and (5) the mixt. formed is incubated at -20 deg.C for 14 hours and the precipitate collected by centrifuging.
