Bacterial transformation is commonly used both in commercial applications and research laboratories to transfer one or more genes into a cell. Chemical transformation of protoplasts is one of the most conventional methods for transformation of the genus Bacillus. The outcome of such a transformation is largely related to the extent of protoplastization. Here we optimize a PEG-induced protoplast transformation protocol by altering the lysozyme concentration and the incubation time.
