PLASMID OF BACILLUS SUBTILIS, BACILLUS SUBTILIS CAPABLE OF PRODUCING L-TRYPTOPHAN CONTAINING SAME, AND USE THEREOF

PURPOSE:To obtain a plasmid of Bacillus subtilis useful for formation of a bacterium capable of producing L-tryptophan, by inserting a DNA fragment having Cmr gene (chloramphenicol-resistant gene) of a phage of Bacillus subtilis containing the cmr gene to a plasmid of Bacillus subtilis. CONSTITUTION:A bacterium belonging to the genus Bacillus, capable of producing L-tryptophan, having a plasmid of bacillus subtilis into which a chromosome DNA fragment containing tryptophan operon having eliminated inhibition of tryptophan synthesis by tryptophan and a DNA fragment having Cmr gene of a phage of Bacillus subtilis having the Cmr gene are inserted, is cultivated in a medium. Then, L-tryptophan is collected from the formed culture medium. A carbon source, mineral, etc. can be used as the nutritive medium. L-tryptophan in the culture solution is adsorbed on an ion exchange resin, eluted with ammonia water, and collected by a means such as concentration, etc.
