TREATMENT OF INERT TYPE PROTEIN USING ULTRAFILTRATION MEMBRANE

PURPOSE:To make possible to collect inactive-type protein accumulated in cell as active-type protein by performing ultrafiltration of solubilized solution from crushed substance of cell, performing molecular weight fractionation and making refolding. CONSTITUTION:In a case transformed host bacterium is cultured and useful substance is produced in cell and the useful substance is collected from said cell, said cell is suspended in buffer solution and crushed by homogenizer, etc., then insoluble fraction is collected by centrifuge, etc. Next, said insoluble fraction is brought into contact with protein modifier (preferably guanidine hydrochloride) to insolubilize and resultant aqueous solution is subjected to molecular weight fractionation using ultrafiltration membrane. Thereafter, intensity of protein modifier in the aqueous solution is reduced.
