PREPARATION OF BLOOD-COAGULATING VII FACTOR OR ACTIVE TYPE BLOOD-COAGULATING VII FACTOR

NEW MATERIAL:A monoclonal antibody having a specific binding property to the blood-coagulating VII factor (FVII) or/and the active type blood- coagulating VII factor (FVIIa), the binding property being dependent on the concentration of a divalent metal cation existing therein. USE:A reagent for separating and purifying the FVII and FVIIa. PREPARATION:For example, a fresh frozen plasma is molten and mixed with a BaCl2 aqueous solution. The produced precipitates are recovered and purified by an ion exchange chromatography and a gel filtration method to prepare the pure FVII, which is administered together with an adjuvant into a BALB mouse to immunize the mouse. Splenic cells are collected from the immunized mouse and fused with mouse myeloma cells. The produced hybridoma is screened, cloned and subsequently cultured to prepare the monoclonal antibody. The monoclonal antibody is immobilized on a carrier to prepare an adsorbent, which is brought into contact with a starting material containing the FVII, etc., to adsorb the FVII, followed by eluting the FVII to provide the FVII or FVIIa.
