PURPOSE:To accurately measure an antigen and an antibody even with respect to a hemolytic specimen containing hemoglobin in high concn. by adding peroxidaze and a phenolic or anilin type 'trinda' reagent to a measuring reagent before using the measuring reagent. CONSTITUTION:In immune nephelometry, for example, a buffer solution containing a surfactant, peroxidase and a phenolic or aniline type 'trinda' phenolic reagent and the antigen or antibody to a substance to be examined are added to serum to be examined to be reacted with the serum to be examined and the turbidity of the serum to be examined is measured. As peroxidase, one having high substrate specificity, for example, peroxidase originating from a 'wasabi' is designated. As the 'trinda' reagent, phenol or N-ethyl-N-(2- hydroxy-3-sulfopropyl)-3,5-m-toluidine is designated. As the surfactant, a nonionic surfactant such as polyoxyethylene alkyl ether is designated. By this method, the oxidation of hemoglobin due to peroxide in the surfactant can be prevented.
