POLYMERASE CHAIN REACTION REAGENT COMPOSITION FOR REDUCING NON-SPECIFIC AMPLIFICATION OF NUCLEIC ACID, TEST KIT AND AMPLIFYING AND DETECTING METHOD

A PCR reagent can be encapsulated with an inert, meltable material which will keep the reagent from interaction in unwanted amplification reactions until its reaction is desired in the amplification of a targeted nucleic acid. Various reagents used in PCR can be so encapsulated, but particularly the DNA polymerase coreagents and deoxyribonucleotide-5'-triphosphates. The encapsulating material is a water-impermeable solid at room temperature, but melts at a temperature of at least about 50 DEG C at which primer extension products are generally formed. The encapsulated PCR reagent can be used in various amplification and detection methods to reduce nonspecific amplifications.
