PURPOSE: To obtain a new transformant useful as an expression system for producing a useful polypeptide. ;CONSTITUTION: The transformant (e.g. a yeast SN35A-1PUAET strain) is obtained by integrating an expression unit containing a gene capable of coding a receptor protein ERD2 (or its analog) derived from a yeast capable of receiving a protein, localized in an endoplasmic reticulum and having an endoplasmic reticular retention signal (e.g. an expression plasmid pIVTRPADHYERD2) and (B) an expression unit containing a gene capable of coding the protein, localized in the endoplasmic reticulum and having the endoplasmic reticular retention signal [e.g. a protein disulfide isomerase (PDI) gene] in a coexpressible state on a yeast chromosome. The exemplified transformant is obtained by transfecting an expression vector containing the yeast ERD2 gene into a yeast SN35A-1PU strain prepared by transducing a human PDI expression unit into a ura3 gene locus of a yeast SN35A strain capable of highly secreting human serum albumin (HSA), causing the homologous recombination at the trp1 site on the yeast chromosome and transducing the ERD2 gene thereinto.;COPYRIGHT: (C)1993,JPO&Japio
