PURPOSE:To suppress the influence of a reducing substance in a specimen on the determination of a peroxidase (POD) activity, by adding a reducible color- developing reagent to a specimen and determining the color of a colored compound produced from the color-developing reagent in the presence of specific reagent. CONSTITUTION:A specimen is added with a color-developing reagent liquid containing a reduced coenzyme, an amine or phenolic compound (or naphthol compound), a reducible color-developing reagent and, if necessary, a surfactant. A solution of H2O2 is added to the mixture and made to react at a specific temperature for a specific period. The reaction is terminated with a proper reaction terminator and the color of the reaction mixture is measured to determine the POD activity. In an immunological dyeing method, a specimen is dyed with a similar method, washed and the dyed part is observed.
