PURPOSE:To obtain a 6-deoxymaltooligosaccharide useful as a reagent for assaying an alpha-amylase activity by treating beta-deoxycyclodextrin with a specified cyclodextrinase and further with an exo-type saccharifying enzyme. CONSTITUTION:A new cyclodextrinase having an action of cleaving cyclodextrin to form a maltooligosaccharide derived from the degree of polymerization of glucose of cyclodextrin is produced by culturing a microorganism such as Bacillus sphericus E-244 strain and collecting the product from the culture. 6-Deoxycyclodextrin is treated with this enzyme and, at the same time therewith or thereafter, with an exo-type saccharifying enzyme. In this way, a new 6- deoxymaltooligosaccharide represented by the formula (wherein n is 2-6) is obtained.
