PURPOSE: To perform a high-sensitivity detection without obstructing the reactivity of a hybridization reaction by a method wherein a sugar is bonded to the heterocyclic ring of a dye intermediate, a phosphate group is ester-bonded to the part of the sugar and a nucleic acid intermediate is formed. ;CONSTITUTION: Sugar is bonded to the heterocryclic ring of a dye intermediate used to form one or more dye structures which contain a dye such as an azo dye, an anthraquinone dye, an indigoido dye or the like and a dye selected from a group composed of their derivatives. The sugar-bonded dye intermediate which is obtained is phophorylated, one to three phosphate groups which are obtained from an inorganic phasphate group and its derivative are ester-bonded to the part of the sugar, and a monoculeotide-resembled dye intermediate is obtained. Then, the mono-nucleotide-resembled dye intermediate introduced into a nucleic acid probe, and a nucleic acid probe intermediate is formed. In addition, a dye intermediate part with which the nucleic acid probe intermediate is provided is made to be dye, a dye structure is formed, and a dye-labeled nucleic acid probe is obtained.;COPYRIGHT: (C)1995,JPO
