PURPOSE: To obtain a new dye capable of binding with a protein such as an antibody and used for the preparation of a labeled antibody useful for the detection of an antigen by an immunochromatographic technique by succinimidylating the new carboxylic acid derivative of a cyanine-based compound. CONSTITUTION: This new cyanine-based carboxylic acid derivative expressed by formula II is obtained by reacting hydrazinobenzenesulfonic acid with isopropyl methyl ketone, then reacting a halogenized alkyl acid to give a carboxyalkylindoleniumsulfonate expressed by formula I (M is H or an alkali metal; n is 1 4; X is a halogen), then dissolving into pyridine after forming its potassium salt, reacting with an orthoformic acid ester while refluxing under an argon gas flow. Next, by reacting this compound with hydroxysuccinic imide in dimethylformamide in the presence of a condensation agent, a cyanine-based labeling pigment expressed by formula III, capable of binding with a protein such as an antibody to give a highly water soluble cyanine labeled protein, is obtained.
