PURPOSE: To obtain a reduction-denatured serum albumin for a treponeme antigen- sensitizer and a determinator of a treponeme antibody, etc., by performing a chemical modification inhibiting rebonding into an intermolecular S-S bond to a SH group generated by a reduction cleavage of the intramolecular S-S bond. ;CONSTITUTION: A serum albumin (e.g. bovine serum albumin) is dissolved in 10mM of diethanolamine buffer solution (pH11) and a reductant such as 2-mercaptoethanol is added to the resultant solution and reduction-treated by reacting at 37°C for 2 hours, a SH group is formed by cleaving an intramolecular disulfide bond, an iodoacetic acid in two times mol of 2-mercaptoethanol is added to the reacted solution and held at a room temperature for 15min. Then, at least one of generated SH group is subjected to a chemical modification capable of inhibiting rebonding into the intramolecular disulfide bond, thus a conjugated substance with a treponeme antigen is carried on the surface of an insoluble carrier to afford the objective reduction- denatured serum albumin useful for a treponeme antigen-sensitizer and a determinator of a treponeme antibody.;COPYRIGHT: (C)1996,JPO
