Purification method of 69000 dalton antigenic protein from Bordetella pertussis

PURPOSE: To obtain the subject antigenic protein in a commercial scale having a specific molecular weight and useful for the production of a vaccine by inactivating Bordetella pertussis cells by a bacteriostatic agent, extracting an outer membrane protein and then purifying by means of a dye ligand chromatography. ;CONSTITUTION: This method for purifying a 69000 dalton antigenic protein from Bordetella pertussis, is obtained by adding a bacteriostatic agent beforehand for inactivating the growth culture material of the Bordetella pertussis (by adding 0. 02 % thimerosal and treating at 37-10°C for 12-18 hr), suspending the cultured material in a buffer solution for incubating to isolate the protein, extracting the protein by plural times at 60-90°C, and purifying the extract by means of a dye ligand chromatography and a chromatofocusing to obtain an outer membrane protein having approximately 69000 dalton molecular weight.;COPYRIGHT: (C)1991,JPO
