The present invention provides a method and a multi-gene assemblyvector system having an acceptor vector and two donor vectors for effectivetransfer of multiple genes into a single vector. Using the method and thevectorsystem disclosed in this invention, multiple cycles of DNA recombination basedon a recombination system can be repeatedly carried out by alternate use ofthetwo donor vectors together with the acceptor vector for recombination. Thevector system and the method are used to construct any genetic engineeringvector that contains multiple genes or DNA fragments of interest for transferandexpression of the genes in different cells, tissues, and organisms.
