PROBLEM TO BE SOLVED: To obtain an oil or fat which contains (n-3) docosahexaenoic acid(DHA) and (n-6) docosapentaenoic acid(DPA) in high concns. and eicosapentaenoic acid(EPA) in a low concn. by culturing the strain SR21 of the genus Schizochytrium capable of producing (n-6) DPA in a medium and extracting an oil or fat from the culture product. SOLUTION: A strain having the same or substantially the same mycological properties as those of the strain SR21 accumulates an oil or fat in an amt. of at least 10wt.% based on dry fungi. The oil or fat contains 6-11wt.% (n-6) DPA, 25-45wt.% (n-3) DHA, and 0.5wt.% or lower EPA based on the total fatty acid. The medium is prepd. by using natural or artificial seawater. A carbon source is used in a concn. of 20-120g/l, and other necessary ingredients are added. The medium is adjusted to a pH of 4.0-6.5 and is sterilized in an autoclade. The fungi is cultured at 10-35 deg.C for 3-7 days by air-flow agitation culture, shake culture, or standing culture.
