PROBLEM TO BE SOLVED: To obtain raffinose synthetase having activity for producing raffinose from sucrose and galactinol, capable of changing a content of oligosaccharide belonging to a raffinose group in plants, and useful in e.g. food materials. SOLUTION: This synthetase has activity for producing raffinose from sucrose and galactinol and has the following physicochemical properties; action and substrate specificity, to produce raffinose from sucrose and galactinol; optimum pH, 6-8; optimum temperature, 35-40 deg.C; molecular weight, 75-95 kDa (gel filtration); 90-100 kDa (polyacrylamide electrophoresis); inhibition, inhibited by iodoacetamide, N-ethylmaleimide and myoinositol. This raffinose synthetase is a protein (A) having an amino acid sequence expressed by formula I or II or (B) having an amino acid sequence wherein one or several amino acid(s) is (are) substituted, deleted, inserted, added or inverted in the amino acid sequence of formula I or II, and has activity for producing raffinose from sucrose and galactinol.
