PURPOSE: A mixed culture of rice bran, Aspergillus oryzae, and Bacillus subtilis, and a preparation method thereof are provided to produce a Koji-substitute zymogen. CONSTITUTION: A method for preparing a mixed culture of rice bran, Aspergillus oryzae, and Bacillus subtilis comprises the following steps of: steaming rice bran at the temperature of 100 ~ 110C for 10-15 minutes; inoculating 0.1-0.2%(w/w) Aspergillus oryzae to the steamed rice bran and cultivating at 32 2C for 12-20 hours; mixing 0.1-0.01%(w/w) Bacillus subtilis, 5-10%(v/w) steamed soybeans, and 30-40%(v/w) water and grinding the mixture; and mixing the rice bran-Aspergillus oryzae culture and wet crushed material of steamed soybean, and cultivating at 40 ~ 45C for 24-30 hours to obtain the mixed culture of rice bran-Aspergillus oryzae-Bacillus subtilis.
