PROBLEM TO BE SOLVED: To establish a technique by which nonspecific amplification is controlled in the nucleic acid amplification in an isothermal amplification reaction to improve amplification accuracy.;SOLUTION: The modified single-stranded DNA-binding protein derived from an extremely thermophilic bacterium has an amino acid sequence exhibiting a function contributing to the improvement of amplification efficiency of a template nucleic acid in an isothermal amplification system using a chain-substituting polymerase.
