The invention relates to a process for isolation and purification of bioactive proanthocynidins possessing anti-inflammatory activity by virtue of its inhibitory action on cylooxygenase -1 and 2 (COX-1 and COX-2).The relief from pain and inflammation Anti-inflammatory Drugs is achieved by blocking production of enzymes cylooxygenase -1 and 2 (COX-1 and COX-2). Under the normal conditions, COX-2 expression is highly restricted; but COX-2 is dramatically unregulated during inflammation and in the rheumatoid arthritis (RA). Hence the inhibition of COX-2 and COX-1 consequently decline of formation of prostaglandins with relief from pain and inflammation. The proanthocyanidins under patent was isolated from the leaves of Anogeissus pendula, a very common tree in deciduous forest of Bundelkhand region of Uttar Pradesh and nearby states and is much popular for fuel wood and fodder in lean periods. It was isolated by simple process of extraction with aqueous acetone and purified by paper chromatography using water as eluting solvent avoiding expensive organic solvents and costly techniques like High Performance Liquid Chromatography (HPLC), Centrifugal partition chromatography (CPC) and gel-permeable chromatography (GPC)etc. The final purification was done by paper chromatography enhancing the economic viabilityof the process. The proanthocyanidins exhibited excellent anti inflammatory activity by inhibiting cox-2 and Cox-1 enzyme with IC50 35.0µgm and 96.0ngm showing cox-2/cox-l ratio 2.74 very much similar to Diclofen, a popular NSAID on OTC which has COX-2/ COX-1 ratio 2.2. Keeping in view the economic viability of process and potentiality of proanthocyanidin as anti-inflammatory activity it possesses all merits to get patent under IPR.
