The claimed invention relates to a method for detecting, differentiating and quantifying T-cell populations, comprising the following steps: a) ensuring that a patient has a first aliquot of biological fluid in contact with at least one antigen, the biological fluid containing antigen-presenting cells (APC) and T-cells; b) incubating the first aliquot with at least one antigen for a certain period of time; c) detection and differentiation of T-cell populations by detecting at least the first APC marker induced by T-cells of a specific T-cell population in the first aliquot and in the second aliquot of the patient’s biological fluid that was not incubated with at least one antigen using quantitative PCR with reverse transcription in real time (RT-PCR RT); and d) detection and quantification of T-cell populations by determining the ratio of the recognized APK marker in the first aliquot to the second aliquot, as well as a kit for the practical implementation of the claimed method.
