The method involves irradiating sample (12,112) with light beam using radiation source (10,110). The sample is detected at surface of detection device (18,118). The detected spectra are stored as function of location, or angular positions of radiation source and detection device. The detected spectrum is adjusted with respect to stored database spectra. The concentration distribution of substances is correlated as function of depth of sample. An independent claim is included for device for determining concentration distribution of substances in sample.
