The present invention displays a polydimethylsiloxane(PDMS) structure, a cell culture instrument using the same, and a manufacturing method thereof. The PDMS structure comprises: a solid body which contains a PDMS material and an elliptical or spherical cavity. The manufacturing method of the PDMS structure to form a cavity on the solid body manufactured by PDMS comprises steps of: a step of supplying incompatible droplets to the PDMS; and a step of hardening process to form a solid body by hardening the PDMS to form the cavity on the solid body by the space filled by the droplets. According to the present invention, a cell culture instrument can be implemented to simulate the conditions of inner body adequate for cells, and a manufacturing process for the PDMS having a cavity with a smaller entrance than the inner space is facilitated since the amount of culturing solution contained in the spherical or elliptical cavity can be increased.
