The present invention relates to a method for culturing a three-dimensional cell using an agarose-collagen-alginate composite hydrogel scaffold. More specifically, the method comprises the steps of: mixing and culturing a cell in an alginate solution; preventing the separation of the cell by adding a collagen solution in the cultured mixture; and adding and stationing an agarose solution in the mixture including the collagen in order to be gelated. In the method of the present invention, a cell culturing environment which is very similar to the human body is composed, and spheroid and spheroid budding forming functions, cell survival function, growing functions, proliferating functions, differentiating functions, and drug diffusing functions are excellent. Accordingly, the method of the present invention can be effectively used in studying the formation, growth, transition, and death of cancer cells, testing drug sensitivity, diagnosing cancer, discerning treatment effects, developing a technique for predicting prognoses, and developing a treating agent having excellent biocompatibility.
