The present invention relates to novel compositions of therapeutic cyclodextrins having polymeric compounds designed as carriers for small molecule therapeutic delivery and pharmaceutical compositions thereof. In particular, the therapeutic / lifestyle agent is covalently bound to the cyclodextrin-retaining polymer. Such cyclodextrin-retaining polymers, when used in vivo, improve the drug stability and solubility of small molecule therapeutics and reduce toxicity. Moreover, by selecting a variety of linker groups and target ligands, the polymer can modulate the delivery method of the therapeutic agent. The invention also relates to a method of treating a subject with a therapeutic composition as described herein. The invention further relates to a method of conducting a pharmaceutical business comprising manufacturing, licensing and circulating a kit comprising or associated with a polymeric compound as described herein.
