The present invention relates to a sensor for measuring protease activity using auto-inhibitory protein, and a method for measuring protease activity. The method for measuring protease activity according to the present invention can solve various problems, such as difficult standardization shown in the conventional protease activity measurement; can be used as a standard protocol for protease activity measurement due to hardness, sensibility, and high convenience; and can be developed as a disease diagnosis method using therapeutic protease for disease.
