1 - Heat stabilization did not effect the morphology of the stabilized tissue and did not compromise pathological review. 2 - Protease activity, that was determined by degradation of synthetic peptide RHPYFYAPELLFFAK), could be blocked in serum by heat stabilization. 3 - In a bottom-up approach where samples were digested with trypsin, we could not find any clear evidence of degradation within the tested time frames. Thus it was not possible to determine the effect of the stabilization method on the degradation. 4 - If the analysis is targeted on natural-occurring peptides and degradation is supported by temperature and time, we could easily observe an effect due to stabilization treatment.
