The measurement method of protease activity is accurate and sensitive and secures high user convenience. Accordingly, the measurement method of protease activity can be used as a standard protocol for measuring protease activity, and further can be developed into a disease diagnosis method. Recombinant pro-caspase-3 includes: a linker including an amino acid sequence which is cleaved by matrix metalloproteinases (MMP) 2. The linker is at least one selected from the group consisting of GPLGVR, GIETGPLGVRS, GIETGSGPLGVRGS, GIETGGSGPLGVRGGS, and GIETGGGSGPLGVRGGGS.
