The present invention relates to non-naturally occurring nucleotide analogs, containing predominantly hydrophobic nucleotide base analogs, which form non-naturally occurring base pairs during DNA polymerase-mediated DNA replication or RNA-mediated RNA transcription. Thus, non-naturally occurring nucleotide bases can be introduced site-specifically into oligonucleotides (single or double-stranded DNA or RNA), where they can provide site-specific cleavage or can provide a reactive linker that can be functionalized with a high-yielding agent reactions with the primary amino group or with the help of click chemistry with the alkyne group of the linker of a naturally occurring nucleotide base.
