A fatty-acid-binding albumin-drug nanoparticle and a lyophilized preparation thereof. The nanoparticle comprises an oily core, albumin, and a drug, wherein the albumin is bound to fatty acids in the oily core and the drug is bound to the albumin and/or fatty acids. The lyophilized preparation of the fatty-acid-binding albumin-drug nanoparticle has high nanoparticle integrity and stability in an in vitro solution and in vivo blood circulation, drug content in gp60-rich receptor cells or tumor tissues can be significantly increased, and LD50 and MTD of load drugs can be significantly improved.
